16 Bit Windows Subsystem Help

I cannot load many programs due to this error message - C;\system32\autoexec.nt. The system file is not suitable for running MS-DOS and Microsoft Windows applications. choose close to terminate this program.    Something is seriously wrong because it is not the applications. They worked fine before?? Help???
