Editing AUTOEXEC.BAT

I edit a line in the AUTOEXEC.BAT file to say:
LOADHIGH=C:\WINDOWS\COMMAND\MSCDEX.EXE /D:MSCD001
This and another line in CONFIG.SYS are added to get my CD-ROM drive to read in dos as drive E:\. It worked once, but I restarted my computer and ever since then it hasnt worked. In fact, everytime I restart my computer, that line is not in AUTOEXEC.BAT. I even re-type it and restart the computer in DOS mode and it still does not read E:\ as my CD-ROM drive. Can somebody please help me with this problem. ???
