What Programs Generate What files

Is there anyway to (1) tell what programs are running in the background? And (2) What program is generating what files?
Example: I was trying to remove temp, bak and other files with G-Lock temp cleaner and get the error message, either the file is in use by another program or another user is using the file? Tho I am the only one on the computer?
